5-Bromo-1-pentene (CAS 1119-51-3) for Chemical Sourcing: A Terminal Alkene with Primary Bromide Dual Functionality
5-Bromo-1-pentene (CAS 1119-51-3), also known as 4-pentenyl bromide, is an aliphatic bromoalkene with molecular formula C₅H₉Br and molecular weight 149.03 g/mol [1]. The compound features a terminal C=C double bond at the 1-position and a primary bromide at the 5-position, providing two chemically orthogonal reactive sites in a single building block [2]. This dual functionality enables sequential transformations that are inaccessible with simple alkyl bromides or unfunctionalized alkenes, making it a versatile intermediate in organic synthesis, pharmaceutical development, and polymer chemistry .
5-Bromo-1-pentene Procurement: Why In-Class Bromoalkene Analogs Cannot Be Directly Substituted
While bromoalkenes as a class share the general feature of combining alkene unsaturation with a bromine leaving group, structural differences in chain length and substitution position critically determine both physical handling properties and synthetic outcomes. The primary bromide in 5-bromo-1-pentene undergoes nucleophilic substitution with minimal steric hindrance, while the terminal alkene participates in addition and cross-coupling reactions with predictable Markovnikov/anti-Markovnikov selectivity [1]. Substituting a shorter-chain analog such as 4-bromo-1-butene (CAS 5162-44-7) introduces a molecular weight differential of approximately 14 g/mol (135.00 vs 149.03) and a corresponding reduction in carbon scaffold length, which directly alters product lipophilicity, chain extension capacity, and the spatial arrangement of downstream functional groups [2]. The following quantitative evidence establishes where 5-bromo-1-pentene demonstrates verifiable differentiation from its closest structural analogs.
5-Bromo-1-pentene Technical Evidence: Quantitative Differentiation Data for Sourcing Decisions
Boiling Point and Density Differentiation of 5-Bromo-1-pentene versus Unsubstituted 1-Pentene
The presence of the bromine substituent in 5-bromo-1-pentene substantially increases intermolecular forces relative to unsubstituted 1-pentene, resulting in a boiling point elevation of approximately 96-97°C and a density increase of approximately 0.62 g/mL . This physical property differential directly impacts distillation separation efficiency and solvent compatibility in downstream processing. The elevated boiling point of 5-bromo-1-pentene places it in a distinct volatility class (flash point 30°C, UN1993 Class 3 flammable liquid) compared to its unsubstituted analog [1].
Molecular Scaffold Length Differentiation: 5-Bromo-1-pentene (C5) versus 4-Bromo-1-butene (C4)
When selecting a terminal bromoalkene building block for chain extension or scaffold construction, the C5 framework of 5-bromo-1-pentene provides an additional methylene unit relative to the C4 analog 4-bromo-1-butene (homoallyl bromide, CAS 5162-44-7) [1]. This additional carbon translates to a molecular weight increase from 135.00 g/mol to 149.03 g/mol (14.03 g/mol difference) and an increase in rotatable bond count from 2 to 3 [2]. For applications requiring specific spatial separation between the reactive termini—such as macrocycle formation, cross-linking agents, or telechelic polymer precursors—this C5 framework provides critical geometric flexibility that the shorter C4 analog cannot deliver.
Quantitative Synthesis Yield: 5-Bromo-1-pentene via 1,5-Dibromopentane Dehydrobromination
A documented large-scale synthesis of 5-bromo-1-pentene from 1,5-dibromopentane achieves 80.1% yield under optimized conditions (140°C, 4 h, hexamethylphosphoric triamide in DMF) with final product content of 99% after rectification [1]. This industrially scalable route leverages the readily available and cost-effective starting material 1,5-dibromopentane, establishing a reproducible synthetic pathway that supports consistent bulk supply. In contrast, alternative syntheses using 4-penten-1-ol with CBr₄/PPh₃ or PBr₃ typically operate at smaller laboratory scale and require more expensive reagents [2].
5-Bromo-1-pentene in Stereoselective Synthesis of 7α-(3-Carboxypropyl) Estradiol
5-Bromo-1-pentene is a critical starting material for the stereoselective synthesis of 7α-(3-carboxypropyl) estradiol, a key estradiol analog . This specific application exploits the compound’s C5 bromoalkene framework to install the 3-carboxypropyl side chain at the 7α-position of the steroid nucleus with defined stereochemistry. The required spatial geometry—a five-carbon chain with terminal bromine adjacent to the steroid core—cannot be replicated by shorter bromoalkenes such as 4-bromo-1-butene or 3-bromo-1-propene, which would install chains of insufficient length, nor by longer homologs that would introduce unwanted conformational flexibility or altered pharmacokinetic properties [1].

Synthesis
1,5-Dibromopentane (100 Kg), N, N-dimethylformamide (100 Kg), and hexamethylphosphoramide (6.2 Kg) were stirred and mixed, and then heated in an oil bath at 140°C for 4 h. Atmospheric distillation collects the fractions, and the collected fractions are washed twice with 100 Kg brine. The organic liquid is distilled to obtain product 5-Bromo-1-pentene.
